5-Amino-1MQ

5-Amino-1MQ (5-amino-1-methylquinolin-1-ium) is a small-molecule quaternary quinolinium cation, most commonly handled as its iodide salt (C10H11IN2). It is not a peptide; its permanent positive charge and compact aromatic structure make it a membrane-permeable, substrate-like ligand of its target enzyme.

5-Amino-1MQ emerged from structure–activity studies of quinolinium-based inhibitors of nicotinamide N-methyltransferase (NNMT), the enzyme that transfers a methyl group from S-adenosylmethionine (SAM) to nicotinamide to form 1-methylnicotinamide. It has been characterised as a selective NNMT inhibitor with low-micromolar potency and is used in cell-based studies of the NAD+ salvage pathway, SAM-dependent methylation balance and adipocyte lipogenesis. It has also been investigated in diet-induced mouse models examining adipose-tissue NNMT activity and metabolic parameters.
