Molecular profile
Ipamorelin is a synthetic pentapeptide with the sequence Aib-His-D-2-Nal-D-Phe-Lys-NH2. It incorporates the non-proteinogenic residues 2-aminoisobutyric acid (Aib) and D-2-naphthylalanine, and was developed from the GHRP-1 series of growth hormone-releasing peptides as an agonist of the ghrelin receptor (GHS-R1a).
In its original pharmacological characterisation, ipamorelin released growth hormone from primary rat pituitary cells with potency and efficacy similar to GHRP-6, and antagonist profiling indicated action through a GHRP-type receptor. GH release was confirmed in vivo in rats and swine, and in swine it did not raise ACTH or cortisol beyond the levels seen with GHRH stimulation, which is why it is described in the literature as a selective growth hormone secretagogue. It has since been used in rodent studies of the GH/IGF-1 axis, including work on longitudinal bone growth in rats, and as a reference ligand in ghrelin-receptor pharmacology.
