GLP-1SG

GLP-1SG contains semaglutide, a synthetic 31-residue acylated analogue of glucagon-like peptide-1, GLP-1(7-37). Relative to the native sequence, Ala8 is replaced by 2-aminoisobutyric acid (Aib), Lys34 is replaced by Arg, and Lys26 carries an 18-carbon fatty diacid attached through a gamma-glutamic acid and two 8-amino-3,6-dioxaoctanoic acid (OEG) spacers.

The medicinal-chemistry programme behind these modifications has been published in detail: the Aib substitution confers resistance to dipeptidyl peptidase-4, while the lipidated side chain was optimised to balance GLP-1 receptor potency against high-affinity albumin binding and an extended plasma half-life in animal studies. Semaglutide is widely used as a reference GLP-1 receptor agonist in studies of receptor activation and cAMP signalling, incretin pharmacology, glucose homeostasis and energy balance in rodent models, and peptide half-life extension strategies.
