Molecular profile
CJC-1295 (no DAC), also called modified GRF(1-29) or tetrasubstituted GRF(1-29), is a synthetic 29-amino-acid, C-terminally amidated analogue of the N-terminal fragment of growth hormone-releasing hormone (GHRH/GRF). It carries four substitutions relative to GRF(1-29)NH2: D-Ala2, Gln8, Ala15 and Leu27. It lacks the maleimidopropionyl-lysine drug affinity complex (DAC) that allows the full CJC-1295 construct to conjugate to albumin.
Native GRF is rapidly cleaved at its N-terminus by a plasma dipeptidyl aminopeptidase to an inactive (3-44) product, which prompted the design of D-Ala2-substituted analogues with improved enzymatic stability. The tetrasubstituted GRF(1-29) scaffold was characterised in rat studies of GRF-receptor activation at the anterior pituitary, where it also served as the peptide backbone of the albumin-binding CJC-1295 bioconjugate. It is used in research on GHRH-receptor pharmacology, cAMP signalling in somatotrophs and the dynamics of pituitary GH secretion.
