SNAP-8 (acetyl octapeptide-3; Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp-NH2) is a synthetic, N-acetylated and C-terminally amidated octapeptide. Its sequence reproduces residues 12–19 of the N-terminal region of SNAP-25 (synaptosomal-associated protein of 25 kDa), extending the SNAP-25-derived hexapeptide acetyl hexapeptide-3 (Ac-EEMQRR-NH2) by two residues.
SNAP-25 is a core component of the SNARE complex that drives Ca2+-dependent vesicle fusion and exocytosis. Peptides patterned on its N-terminal SNARE motif are studied as SNARE mimetics that can compete with native SNAP-25 during complex assembly; the parent hexapeptide was reported to interfere with SNARE complex formation and to reduce Ca2+-dependent catecholamine release from permeabilised chromaffin cells. SNAP-8 is used in in vitro studies of SNARE assembly, regulated exocytosis and structure–activity relationships of SNAP-25 mimetic peptides.
