Molecular profile
Cagrilintide is a synthetic, long-acting analogue of the pancreatic peptide hormone amylin. The 37-residue peptide keeps the amylin-type Cys2–Cys7 disulfide bridge and ends in a C-terminal prolinamide. It carries helix-stabilising substitutions, including N14E and V17R, introduced to reduce amyloid-like fibrillation, and is N-terminally acylated with a C20 fatty diacid through a γ-glutamic acid linker.
Cagrilintide is characterised as a non-selective agonist of the calcitonin receptor (CTR) and of the three amylin receptors (AMY1, AMY2 and AMY3), which form when CTR pairs with receptor activity-modifying proteins (RAMPs). Its design and pharmacological characterisation were described in a medicinal-chemistry development study. Later structural and molecular-dynamics work compared its receptor-bound conformation and receptor-complex dynamics with those of other calcitonin-family peptides. It is used in research on amylin-receptor pharmacology and the central regulation of food intake and energy balance.
