Mazdutide

Mazdutide (LY3305677, IBI362) is a synthetic 34-residue analogue of oxyntomodulin that acts as a dual agonist of the glucagon-like peptide-1 receptor (GLP-1R) and the glucagon receptor (GCGR). The sequence carries 2-aminoisobutyric acid (Aib) at position 2, a C-terminal glycinamide, and a lysine at position 20 acylated with a C20 fatty diacid through a γ-glutamic acid and two 8-amino-3,6-dioxaoctanoic acid (OEG) spacers, a half-life-extension motif associated with reversible albumin binding.

Mazdutide is used in studies of combined incretin and glucagon receptor signalling in glucose and energy homeostasis. In db/db mice it has been investigated in studies of cognitive dysfunction linked to metabolic impairment, where behavioural testing was combined with transcriptomic, proteomic and metabolomic profiling and compared with a GLP-1R-selective agonist.
