Molecular profile
GHRP-6 (growth hormone-releasing hexapeptide; His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) is a synthetic, C-terminally amidated hexapeptide containing two D-amino acids. It emerged from structure–activity work on met-enkephalin analogues and is one of the earliest characterised synthetic growth hormone secretagogues (GHS).
GHRP-6 is an agonist of the growth hormone secretagogue receptor type 1a (GHS-R1a), the G protein-coupled receptor later identified as the endogenous ghrelin receptor. Early studies described its specific GH-releasing activity in rat pituitary preparations in vitro and in vivo, and synthetic secretagogues of this class guided the subsequent cloning of GHS-R1a from pituitary and hypothalamus. It remains a widely used tool compound in studies of ghrelin-receptor signalling, somatotroph function and hypothalamic GHS binding sites in rodent and cell-based models.
