Molecular profile
Sermorelin (GRF(1-29)NH2) is a synthetic 29-amino-acid peptide that reproduces the N-terminal 1–29 sequence of human growth hormone-releasing hormone (GHRH, 44 residues), with a C-terminal amide. It is generally regarded as the shortest fully functional fragment of GHRH.
GHRH was first isolated and sequenced from a human pancreatic tumour. Subsequent structure–activity work in rats showed that the 1–29 amide fragment was as potent in vivo as the full-length peptide. Sermorelin acts as an agonist at the GHRH receptor on anterior pituitary somatotrophs, a class B G protein-coupled receptor that signals mainly through Gs and cAMP. It is used as a reference GHRH-receptor agonist in studies of somatotroph signalling, the dynamics of GH secretion and GHRH structure–activity relationships. It is also the parent sequence for modified analogues such as tetrasubstituted GRF(1-29).
