GLP-2TZ

GLP-2TZ contains tirzepatide (LY3298176), a synthetic 39-residue peptide built on the glucose-dependent insulinotropic polypeptide (GIP) sequence that acts as a dual agonist at the GIP and glucagon-like peptide-1 (GLP-1) receptors. It contains 2-aminoisobutyric acid (Aib) at positions 2 and 13, a C-terminal amide, and a C20 fatty diacid linked to Lys20 through a gamma-glutamic acid and two AEEA spacers.

In receptor-pharmacology studies tirzepatide has been characterised as an imbalanced dual agonist, binding the GIP receptor with an affinity comparable to native GIP but the GLP-1 receptor more weakly than native GLP-1, and as a biased GLP-1 receptor agonist that favours cAMP generation over beta-arrestin recruitment. Cryo-EM structures of tirzepatide bound to GIPR and GLP-1R have been solved, and the compound is widely used in cell-based and rodent research on incretin receptor signalling and glucose homeostasis.
