Molecular profile
GLP-3RT contains retatrutide (LY3437943), a synthetic 39-residue peptide that acts as a single-molecule agonist at the glucose-dependent insulinotropic polypeptide (GIP), glucagon-like peptide-1 (GLP-1) and glucagon receptors. Its sequence includes two 2-aminoisobutyric acid (Aib) residues and an alpha-methyl-L-leucine, ends in a C-terminal amide, and Lys17 carries a C20 fatty diacid attached through a gamma-glutamic acid / AEEA linker.
Retatrutide is used in receptor-pharmacology research on multi-agonist incretin and glucagon signalling. Cryo-EM structures of retatrutide bound to GLP-1R, GIPR and GCGR have been reported, giving a structural basis for how one peptide engages all three class B G-protein-coupled receptors. It has also been studied in diet-induced mouse models and in rodent research on glucose regulation and energy balance.
